Type 1 diabetes genetic susceptibility markers and their functional implications

چکیده
against islet cell autoantigen (ICA). More than 90% of individuals with T1D test positive for at least one such autoantibody, and the presence of autoantibodies is used to identify the relatives of patients who are at increased risk of the disease [2-4]. Although genetic factors are strong determinants of the disease, more than 90% of individuals who present with newonset dia betes do not have a relative with T1D [3]. The overall risk of T1D for the siblings of T1D patients is approximately 6%, which is 15 fold higher than in the general population [5]. The most important susceptibility alleles are found within the human leukocyte antigen (HLA) region, which has an odds ratio for disease of approximately 6.8 [6]. Single Nucleotide Polymorphism (SNP) in the Adiponectin Gene and Cardiovascular Disease
Dear Editor, The recent article by Mohammadzadeh et al.[1] on the latest issue of this Journal showed that the T allele +276G/T SNP of ADIPOQ gene is more associated with the increasing risk of coronary artery disease (CAD) in subjects with type 2 diabetes. Adipocytes were described in myocardial tissue of CAD patients and their role recently discussed[2,3]. Susceptibility to CAD by polymorp...
متن کاملEvidence for a novel type 1 diabetes susceptibility locus on chromosome 8.
Type 1 diabetes results from a combination of genetic susceptibility and environmental exposures. Susceptibility loci other than HLA and the insulin gene remain to be identified to account for the degree of familial clustering observed in this disorder. Early genome-wide scans provided suggestive evidence of linkage on chromosome 8q, prompting detailed analysis of this region. A total of 20 mic...
متن کاملPhysical and genetic mapping of IDDM8 on chromosome 6q27.
Genome-wide mapping studies have provided evidence of a type 1 diabetes susceptibility gene (IDDM8) that is located on chromosome 6q27. However, association studies of IDDM8 have so far been negative. The purpose of this investigation was to determine a linkage disequilibrium (LD) map in the chromosome 6q27 region and to better localize IDDM8. A physical map of nearly 1 Mb containing the chromo...
متن کاملHLA Class II Alleles Susceptibility Markers of Type 1 Diabetes Fail to Specify Phenotypes of Ketosis-Prone Diabetes in Adult Tunisian Patients
We aimed to characterize the different subgroups of ketosis-prone diabetes (KPD) in a sample of Tunisian patients using the Aβ scheme based on the presence or absence of β-cell autoantibodies (A+ or A-) and β-cell functional reserve (β+ or β-) and we investigated whether HLA class II alleles could contribute to distinct KPD phenotypes. We enrolled 43 adult patients with a first episode of ketos...
متن کاملDiabetes mellitus in a young Amazon Indian child.
CONTEXT Although type 2 diabetes has been described among American Indian children, no case of type 1 diabetes has been reported in the literature. CASE REPORT We report the first case of diabetes in a South American Indian child from the tropical rainforest, who was positive for IA2 autoantibodies and genetic markers of susceptibility to type 1 diabetes, but also demonstrated residual beta c...
